OPTIMIZATION OF ALKALINE PROTEASE FROM ALKALIPHILIC ACTINOMYCETE ISOLATED FROM ALKALINE SOIL OF BHUJ, GUJARAT, INDIA

Abstract

Three alkaliphilic actinomycetes were isolated from alkaline soil of Bhuj, Gujarat, India. All the three isolates were able to grow luxuriously at p1-I 10. Alkaline protease production was optimal in exponential phase of growth. The maximum enzyme secretion was observed in presence of Glucose (1%, w /v) and peptone (0.5%, w /v) as the sole source of carbon and nitrogen respectively. The growth and enzyme production was found to be suppressed with NI-11C1 (0.5%, w/v) and Urea (0.5%, w/v). Cations Ca, Mn++ (20 mM) repressed enzyme production significantly with higher growth. Whereas, Mg ++ (20 mM) found have lesser repressed effect on enzyme production with slight enhancement in growth. Whey supplemented with K2HPO4 (1%, w/v) enhanced the protease activity by two fold.
