COMPARISON BETWEEN ZIEHL NEELSEN AND AURAMINE-O STAINING FOR DETECTION OF ACID FAST BACILIRITU KANSAL, ANITA PANDEY, ASHISH K. ASTHANA AND R.K. AGRAWALAbstract Microscopic examina lion of respiratory specimens for acid-fast bacilli (APB) plays a key role in the initial diagnosis of tuberculosis as well as for monitoring of an titubercula r treatment. The objective of this study was to compare the sensitivity of Ziehl Neelsen (ZN) and Fluorescent staining (auramine 0) in the detection of Mycobacteria. A total of 250 early morning sputum specimens were collected from Clinically suspicious cases of pulmonary tuberculosis at C.S.S.H., Subharti Medical College, Meerut, India. These samples were processed as per CDC guidelines. One hundred ninty five cases were found to be Positive for AFB. Out of which 85 were stained satisfactory with ZN and 110 with AO staining method. Thus, Fluorescent staining method was found to be more sensitive in detecting acid fast bacilli which were missed by ZN method.
