BIRENDRA SINGH, IFFAT ZAREEN AHMA D AND S. SHANTHY

Abstract

Fast growing cultures of Launaea nudicaulis can be achieved through callus culture in Murashige and Skoog medium with 3% sucrose and 0.8% agar, good callus growth was observed in MS medium containing 2.5 ppm 2, 4-D and 0.1 ppm kinetin. These calli can be maintained in MS media of half strength with similar hormonal concentration by regular sub culturing at every 23rd day. Organogenesis was better in MS media containing 2.0 ppm IBA and 0.1 ppm IAA. Cultures incubated in dark showed marked differentiation in comparison to light. Some calli were maintained in MS medium of half strength containing 0.2 ppm IBA and 0.1 ppm IAA after excision of small plantlets, which provides continuous source of micropropagules. Rooting of the shoots was maximum in MS medium supplemented with 2.0 ppm IBA.
