SELECTION AND VALIDATION OF REFERENCE GENES IN GOSSYPIUM HIRSUTUM L. UNDER DROUGHT STRESS BY QUANTITATIVE RT-PCR

Abstract

results, normalization of real-time PCR data is required against one or more suitable reference genes, which displays highly uniform expression in living organisms during various phases of development and under different environmental conditions. We selected and validated the expression stability of six reference genes, including Histone3, GhTUA11, Polyubiquitin, GAPDH and two Ubiquitin genes in drought stressed and well watered cotton samples by qRT-PCR. Their expression varied considerably in different tissue samples analyzed. By using NormFinder and geNORM software package the genes, GhTUA11 and Polyubiquitin exhibited most stable genes in cotton leaf and root samples grown under drought stress and control (well watered) conditions. Thus, GhTUA11 and Polyubiquitin may be used as reliable reference genes for gene expression analysis in G. hirsutum under drought stress condition.
