MYCOBACTERIUM TUBERCULOSIS DETECTION FROM DUST AND WATER SAMPLE IN HOUSE OF BTA (+) LUNG TUBERCULOSIS PATIENT THROUGH POLYMERASE CHAIN REACTION METHOD IN MALANG

Abstract

Tuberculosis is one of health problems in the worldwide and one of the endemic countries is Indonesia. The prevalence of tuberculosis in East Java is considered as one of the highest in Indonesia, for instance, the prevalence in Malang is 0.4% with 60% of it are detected as BTA (+). This study was conducted to investigate M. tuberculosis bacteria in dust and water sample that are found surrounding the home of tuberculosis patients in Sukun district in Malang. Samples were analyzed using Polymerase Chain Reaction (PCR) with Pt 3 and Pt 6 primers. Pt 3 and Pt 6 located in the 188 bp as well as the primary Tr 8 and Tr 9 is located at 157 bp. Bacteria were cultured in Middle Brook medium and Lowenstein- Jensen. There are 5 samples (27.8%) detected as positive and the rest of it or 13 samples are negative. Positive samples PCR band show similarities with band 100-200 bp. Furthermore, there are 6 water samples (33%) that detected as positive with the appearance of 188 bp bands. These results indicate the transmission risk in Sukun area so there will be more studies required.
