CYTOTOXICITY STUDY OF FREEZE-DRIED BOVINE BONE XENOGRAFT IN HUMAN BONE MARROW MESENCHYMAL STEM CELL

Abstract

Xenograft materials can be obtained from cow bone or called bovine-derived bone xenograft (FDBBX). Freeze-dried bovine bone xenograft (FDBBX) is made from bovine cortical bone which was rich of growth factors in freeze drying manufacturing process. Biocompability of this graft material still needs to be proven. The aim of this study is to evaluate cytotoxicity of FDBBX applied in human bone marrow mesenchymal stem cells (hBM-MSC) culture. hBM-MSCcultures were divided into experimental and control groups. Each experimental group has 3different concentration of FDBBX conditioned medium (2.5%, 5%, and 10%) while control group consist of basic medium. The amount of viable cells were measured using MTT Assay in both groups after 24 and 72 hours of cells culture. Both control and experimental groups showed the amount of cell viability was above 60%. The highest number of viable cells was shown in 2.5% FDBBX’s group. There was no statistically difference between 24 and 72 hours of cells culture (p<0.05). Freeze-dried bovine bone xenograft is not cytotoxic with optimum concentration of FDBBX conditionedmedium being at 2.5%.
