Abstract

The aim of this study is to investigate the distribution of ACE gene I/D polymorphism and its association with steroid responsiveness in Iraqi children with nephrotic syndrome (NS). Fifty children with NS were divided into four groups: 14 steroid sensitive infrequent relapse (SSIFR), 12 steroid sensitive frequent relapse (SSFR), 9 steroid resistance (SR) and 15 newly diagnosis (ND)compared to twenty-five unrelated healthy children. Each individual genotype was determined using PCR amplification of extracted genomic DNA and allele distribution based on size of the PCR fragments and ACE genotypes DD, ID and II were compared between different groups. The frequency distribution of the DD genotype was significantly elevated in patients with NS compared to control subjects (P=0.0001). Our results showed that NS is associated with higher incidence of DD genotype, especially in SR patients. Angiotensin II may be involved in part in the response to steroid treatment in children with NS.
