DETERMINATION OF THE OPTIMUM CONDITIONS OF LACCASE PRODUCED FROM LOCAL ISOLATE OF CITROBACTER FREUNDII USING SUBMERGED FERMENTATION

Abstract

In the current study seven different bacterial isolates, comprising Klebsiella pneumonia, E. coli, Citrobacter freundii, Burkholderia mallei, Enterobacter aerogenus, Citrobacter amalonaticus and Pseudomonas putida were previously isolated from the waste active sludge tank and radiated by heterogeneous photocatalysis (UV+TiO2) using a lab scale photoreactor. Then the isolates were identified and subjected to a quantitative screening process using submerged fermentation to select the best isolate for laccase production. Among these isolates, Citrobacter freundii was chosen as it had the highest enzyme productivity (6083.3 U/mg protein). The optimum conditions of laccase produced by selected isolate were obtained in themineral salt medium (MSM) supplemented by 1% glucose as the best carbon source and 3% urea as the nitrogen source in pH 6.0 at 40 °C after 24hrs of incubation.
