DIRECT ADVENTITIOUS SHOOT BUD REGENERATION AND EX VITRO ROOTING IN LEAF EXPLANTS OF FEMALE SIMMONDSIA CHINENSIS (LINK) SCHNEIDER- AN IMPORTANT INDUSTRIAL SHRUB

Abstract

Simmondsia chinensis (Link) Schneider is known as the Gold of Desert, an important industrial plant. High frequency in vitro adventitious shoot regeneration and ex vitro rooting from leaf explants of female S. chinensis has been developed. The highest (90.8±1.16%) response of explants was obtained on Murashige and Skoog's (MS) medium containing 1.0 mg/L thidiazuron (TDZ) with maximum number (4.57±1.34) of shoots and (2.83±0.25 cm) shoot length. The presence of TDZ in culture medium has great influence on direct shoot bud induction than (indole-3-butyric acid) IBA and 6-benzylaminopurine (BAP). The multiplication and elongation of shoots were obtained on MS medium containing 0.5 mg/L BAP and 20 mg/L adenine sulphate (AdS). The micro propagated shoots were rooted ex vitro after treatment of 500 mg/L IBA for 3 min. The hardened and acclimatized plantlets were transferred successfully to the field without morphological variations. This protocol is simple and efficient for large scale production of true-to-type plants and helps in commercial establishment of this plant.
