IDENTIFICATION OF PHYTOPATHOGENIC BACTERIA OF GENUS PSEUDOMONAS USING THE REAL-TIME PCR METHOD

Abstract

In increasing the yield of grain and legume crops, an important role is played by their protection from diseases that often result in a significant reduction of crop production, deterioration of quality, and sometimes harvest failure. One of the reasons of harvest failure is the presence of pathogenic bacteria species. Such pathogenic bacteria species include Pseudomonas syringae, Pseudomonas pisi Sackett, and Pseudomonas holci Kendrick. Detection and taxonomic identification of the pathogenic bacteria is important, and almost indispensable not only in agriculture but also in other areas, such as food safety and environmental monitoring. In this research, these bacteria were identified using the method based on real-time PCR, with a matching pair of primers and a probe, which allowed identifying bacteria of genus Pseudomonas wide spread on the territory of Kazakhstan: Pseudomonas syringae, Pseudomonas pisi Sackett, and Pseudomonas holci Kendrick. Identification of bacteria is based on PCR amplification of DNA fragments using specific primers that have been developed based on the gene of subunit B of DNA gyrase.
