PRODUCTION OF ACIDIC INVERTASE BY PENICILLILIM PERPUROGENUM AND EFFECT OF SUBSTRATE FEEDING

Abstract

Penicillium purpurogen urn was found to produce both extracellular as well as intracellular acidic invertases (13-fructofuranosidases). Optimum extracellular invertase production reached on day 4 and for maximum intracellular enzyme 5 days incubation time was required. Sucrose was used as a carbon source for invertase production. The optimum temperature for invertase activity was at 37°C. The invertase was stable in pH range of 3.5 to 6.0 and had an optimum pH at 4.5. Addition of vitamins to the growth medium has no significant impact on invertase production and supplementation of surfactants completely inhibited the growth of organism. Elevation in invertase production by Penicilliurn purpurogenum was observed when fresh substrate feeding was performed to the cultivation medium during incubation time.
