CHARACTERISATION OF TRANSFERABLE DRUG RESISTANCE PLASMID OF VIBRIO ALGINOLYTICUS INFECTING POST LARVAE OF FRESHWATER PRAWN, MACROBRACHIUM ROSENBERGII

Abstract

The use of wide range of antibiotics to reduce losses associated with vibriosis has resulted in an increased population of multidrug resistant Vibrio strains. In the present study, two Vibrio species viz. V alginolyticus & V. mimicus were isolated from dead post larvae of freshwater prawn, Macrobrachium rosenbergii on the basis of their growth on TCBS agar and reaction to a series of biochemical tests. Among these, V alginolyticus was found to be multid rug resistant like polymyxin-B, ampicillin, sulphadiazine, gentamicin, nalidixic acid. Isolation of plasmid by miniprep plasmid extraction method followed by agarose gel electrophoresis has revealed the presence of a 10 Kb plasmid only in V alginolyticus. Transformation of E. coli DH5“. strain with the 10 kb plasmid resulted in ampicillin resistanv. This concludes that in V alginolytics at least ampicillin resistance property is conferred by the transferable 10 kb drug resistance plasmid. 1993).
