BIOSYNTHESIS OF PIM BY PROMISING NATIVE BACTERIAL ISOLATES OPTIMIZATION OF PROCESS PARAMETERS

P.S. CHANDRASHEKHARAIAH AND K.S. JAGADEESH

Abstract

Out of ten promising PHB accumulating native bacterial isolates, Pesudomonas DJC6 was found to be the most efficient isolate. The process parameters were optimized. Glucose (1%) and ammonium sulphate (0.1%) were found to be the best C and N sources respectively. A C/N ratio of 20:1 and a pH of 7.0 were found to be optimum. Under the optimized conditions, in 3L bench scale digesters, the strain produced a PHB yield of 1.10g/100mL which was marginally higher than the PHB produced by the reference strain, Ralstonia eutropha. It would be very difficult to imagine our lives without plastics. From automobiles to medicine, plastics are utilized in almost every manufacturing industry in the world. About 100 million tons of plastics are produced world wide. Plastics are truly significant in our indutrial society. The disposal of The success in the biodegradable plastic strate largely depends on the development of efficiE polyhdroxyalkanoate producing bacteria. Recent promising PHB synthesizing bacterial isolates frc different environmental samples have been isolat and selected (Cristina et al., 1999)
