CALLUSING, SOMATIC EMBRYOGENESIS AND ORGANOGENESIS IN CAJANL1S CA JAN L.

Abstract

Efficient and reproducible protocol for the micropropagation of the recalcitrant crop Cajanus cayn L. (VBNI) was developed. The combination of auxins (2, 4-D, NAA) with cytokinins (KIN, BAP) was effective for the induction of organogenic callus. Individual concentrations of auxins (IAA, 2, 4-D, NAA ) was effective of the induction of callus, but organogenesis did not occur, Lower concentration of auxins 2, 4-D (0.5 mg/1) and NAA (0.5 mg/1) with cytokinins (BAP, KIN) increased the organogenic potential of hypocotyl, epicotyl, cotyledon and leaf explants. Regeneration of shoot was maximum from cotyledon node explant of MS medium containing BAP. GA3 is vital for shoot elongation. Maximum elongation of shoots from all the explants occurred on MS medium containing GA, (0.5 mg/1) and BAP (1mg/1). IBA (0.5 mg/ 1) was effective for root induction. Induction of embryogenic callus occurred on MS solid medium containing 2,4-D (1.5 mg/1). Maximum induction of somatic embryos was formed on MS Liquid medium containing 2, 4-D (1.0 mg/1). MS medium was effective when compared to other media such as B5, L6, Ms- Bs.
