TRANSFORMING OF GUS GENE IN INDICA RICE CULTIVAR BY AGROBACTERILIM TL1MEFACIENS

Abstract

Genetic transformation of rice mediated by Agrobacterium tumefaciens has been confirmed to japonica and Jhdica rice varieties. Three weeks old scuteilar derived embryogenic caffi of Pusa Basumathi - I were infected with Agrobacterium strain AGL -1 containing P - CAMBIA 1201 vector coding genes for p - Glucoronidase (GUS) enzyme with hygromycin (hpt) resistant marker. Transformants were selected on hygromycin (50mg/L)medium with acetocyringone (100 uM). The efficiency of transformation was improved in addition of acetocyringone during co-cultivation process. The transformation of P - CAMBIA 1201 Vector through Agrobacterium was proved by Gus assay and PCR analysis.
