MTT ASSAY BASED UREAPLASMA UREALYTICUM INFECTION QUANTIFICATION IN PREECLAMPSIA

Abstract

Preeclamptic placental Infection monitor remains vital due to their debilitating nature that leads to adverse maternal and fet;li Preeclamptic Ureaplasma urealvticum infection is largely overlooked from analysis. Difficulty, in the cultivation of this commensalic microbe was also noted. This study analyses the isolation of U. urealyficom 'from the placental chorion of a preeclamptic subject; with different agar medium namely A8 agar, rno agar, Soya peptone agar and A3B agar in a 5% CO, and 100% CO, rich atmosphere. Different cultivation methods namely the spread plate and pour plate technique was followed. 3-(4, 5- Dirriethy1-2-y1)-2,5-Diph.enyl tetrazolium bromide (MTT) assay is used for the first time to identify and quantify the isolate as U. urealyticum. This novel method for U. urealyticurn growth analysis showed high level of significance over the routine growth analysis performed by counting of colonies formed: A8 agar, the commercially available medium depicted a significant variance on internal agar-agar comparison using colony count and a highly significant variance on intrinsic agar-agar comparison with the MIT assay. This is the first report that emphasizes the efficiency of MTT assay, as a routine diagnostic tool for U. urealyticum viability studies.
