HIGH CELL DENSITY CULTIVATION OF ANDROGRAPHIS PANICULATA FOR CELL SUSPENSION CULTURE IN SHAKE FLASK AND BUBBLE COLUMN REACTOR FOR THE PRODUCTION OF CELL BIOMASS AND ANDROGRAPHOLIDE

Abstract

Andrographis paniculata is a well known traditional plant which yields active botanical ingredient andrographolide responsible to treat a variety of ailments. The quality of the drugs is inconsistent due to several factors. To overcome these issues, the aeration rate played an important role in cell growth and constituent substance production. In this paper we investigated the effect of various inoculums sizes (0.55%,1%,2%,3%,4% and 5%) using shake flask and aeration rate (0.166,0.416,0.833,1.25 and 1.66 LPM) using the 2L bubble column bioreactor was used for high cell density suspension cultivation of Andrographis paniculata cells under the optimal culture condition. In comparison of the optimum level of biomass obtained from 4% inoculum size was 12.10g/L and growth ratio was 3.94 respectively. Similarly, various air flow rates with a constant inoculum size in a bubble column reactor were used. An optimum level of biomass was obtained at a flow rate of 1.25 LPM were 13.10 g/L and growth ratio was 3.99 respectively. The cultivation of A.paniculata using bubble column reactor was suitable for large scale production when compared to that with shake flask due to the maximum accumulation of biomass in bubble column reactor
