ISOLATION AND CHARACTERIZATION OF MESOPHILIC LIPASE PRODUCING BACTERIA

Abstract

Bacterial isolates were obtained from sewage water, petrol pump and soil contaminated with groundnut waste on rhodamine olive oil agar (ROA) and Tween 20 agar. Eight morphologically different bacterial colonies named NI1, NI4, S1, S2, S3, S4, MS_1 and OA were picked randomly and maintained on nutrient agar slants. These isolates were subjected to morphological and biochemical tests and were analysed for quantitative enzyme production. When grown on production medium containing castor oil, MS_1 showed highest enzyme activity (21.29 U/mL) followed by S3 (20.76 U/ml) and OA (15.34 U/mL). Media optimization for carbon source, nitrogen source, pH and temperature was carried out. 0.5% castor oil was found out to be best carbon source while 1% ammonium chloride supported maximum growth and lipase production. Optimum bacterial growth was observed at pH 7 and temperature 37°C. The isolate MS_1, which showed best enzymatic activity, was further characterized by 16S rRNA sequencing and was identified as Bacillus subtilis JNT02.
