MULTIPLE SHOOT REGENERATION FROM COTYLEDON WITH HALF EMBRYONIC AXES EXPLANTS OF CHICKPEA (CICER ARIETINUM L.)

Abstract

In the present study, we report successful multiple shoot regeneration from cotyledon with half embryonic (CHA) explants of an Indian chickpea cultivar C 235. Explants were cultured on different shoot induction medium (SIM) containing Murashige and Skoog (MS) medium supplemented with benzylaminopurine (BAP), indol butyric acid (IBA) and naphthalene acetic acid (NAA). Maximum number of shoots per explants was recorded on MS medium containing 1.0 mg/L BAP with 0.05 mg/L NAA. Histological study of regenerating explants showed direct adventitious origin of shoots from the explants. Multiple shoots obtained were elongated in gibberellic acid (GA3) containing medium and were grafted on root stocks prepared from the same cultivar of chickpea.
