Abstract
Molecular characterization of six chickpea genotypes, each differing for Fusarium wilt reaction , was carried out using DNA based molecular markers tagged with disease resistance/susceptibility. In the present study, four molecular markers, one Allele Specific Associated Primer (ASAP) (CS27) and three Sequence Tagged Microsatellite site primers (TA27, TA37 and TA96), were validated. It was found that the ASAP marker, CS27, amplified an allele of 650 bp in susceptible genotype and no amplification was observed in resistant genotypes. At the same time it also confirmed the hybridity of F1s developed by 6×6 diallel mating design. Three STMS markers amplified alleles specific to resistance and susceptibility. The primer TA27 amplified two alleles, one of size 208bp in resistant and other of size 190 bp in susceptible genotype. However, TA37 amplified an allele of size 290 bp in resistant genotypes only. Further, the primer TA96 amplified two bands, each of size 265 bp and 270 bp, in usceptible and resistant genotypes respectively. Therefore, these markers hold their promise in discriminating resistant and susceptible genotypes in our population and hence, can be further used reliably in marker assisted selection for wilt resistance in chickpea.
