MOLECULAR IDENTIFICATION AND OPTIMIZATION OF PROCESS PARAMETERS FOR ANTIMICROBIAL COMPOUND PRODUCTION BY STREPTOMYCES SP.

Abstract

In the present study the molecular identification of S1, an actinomycetes isolate of Bhitarkanika mangrove forest soil of Odisha, India was done. Optimization study of the different cultural parameters on the growth and antibacterial metabolite production by the isolate was carried out by submerged fermentation using shake flask method followed by antimicrobial metabolite extraction using solvent (ethyl acetate) extraction method. The 16S rDNA sequence was deposited in NCBI Genbank under the Accession Number KR401259. The antagonistic activity against the human pathogens was studied by agar well diffusion method using the dried ethyl acetate extract. Enhanced production of the bioactive metabolite by the isolate was observed using starch casein broth under submerged fermentation at 30°C, 1.0% salinity and at an initial pH of 7.5.
