ISOLATION AND IDENTIFICATION OF A CELLULOLYTIC BACILLUS FROM RUMEN OF ACEH’S CATTLE

Abstract

The purpose of this research was to isolate and identify bacteria that produce cellulase enzyme from rumen fluid of aceh cattle. Biodegradation by cellulase producing bacteria found in rumen can be used as source of cellulolytic bacteria which function to degrade feed material with high coarse fiber so as to improve the quality of nutrition and obtain easily digested feeding materials at a cheaper price than the use of commercial cellulase enzymes. Rumen fluid of aceh’s cattle was taken for isolation and screening of cellulolytic bacteria. The FKH_7 isolate showed ± 2.0 cm of clearance zone diameter. Microscopically, this strain was found to be a Gram-positive, Bacillus. Analysis of 16S rRNA and phylogenetic analyses. Homology analysis showed that FKH_7 isolate showed 87% degree of sequence similarity with that Lysinibacillus fusiformis. This study concluded that some potent cellulose degrading bacteria have been identified in rumen of aceh cattle and that has group Lysinibacillus.
