CHARACTERIZATION BY MALDI - TOF MS AND PCR SEQUENCING OF THE FUNGI ASSOCIATED WITH DURUM WHEAT SEED TREATED WITH THE AZOLE FUNGICIDE IN CONSTANTINE (ALGERIA)

Abstract

The main objective of this study is to highlight the fungal species associated with durum wheat seed treated or not with a fungicide azole in order to assess the degree of protection of the seeds after a fungal treatment. The seeds were treated with Tebuconazole 060 FS (Raxil® 060 FS) and the mycoflora was assessed by direct and indirect methods. During the seasons 2014 and 2015, 78 fungal isolates were isolated from treated and untreated seeds. The fungal genera identified by classical identification methods are Aspergillus, Penicillium, Trichoderma, Rhizopus, Botrytis, Alternaria, and Cladosporium. During isolation, the indirect (suspension-dilution) method resulted in a greater number of fungi than blotting (direct method). Characterization of isolated fungal species was determined by PCR sequencing and by mass spectrometry (MALDI-TOF MS). It appears that seeds treated with Raxil® 060 FS show a lower diversity of fungus (31 species) than untreated seed (47 species).
