Abstract
The dot-blot assay is useful for the sero-diagnosis of patients with clinical signs and symptoms consistent with typhoid fever. The advantages of the dotblot assay are that the result can be obtained on the same day allowing a prompt treatment using a small volume of serum without any special laboratory and equipment. Typhoid is caused by Salmonella typhi which can be sero-diagnosed by dotblot assays based on lipopolysaccharides. The role of Salmonella extracted Vi antigen in dot blot ELISA is not very much clear. This study implicates the application of Vi antigen on nitrocellulose membrane and standardization of Dotblot ELISA for its establishment in typhoid diagnosis in future. The Vi-antigen provided specific and sensitive results with rabbit raised Vi antibodies in dotblot assay. This dotblot assay was performed in comparison to the ELISA and the complete standardization of this assay was performed. This study labels to the usage of Vi antigen of Salmonella typhi for establishment of an easy rapid test for detection of IgG and IgM antibodies in typhoid patients.
