MOLECULAR DNA MARKING OF SIBERIAN TOMATO VARIETIES FOR THE PURPOSE OF CERTIFICATION

Abstract

Currently, due to the large varietal diversity and the low-quality seeds at the same time, as well as the violations in seed production, there is a need in genetic identification and development of the methods of molecular diagnostics of plants with subsequent certification of agricultural crops. In this work, the genetic material of the main varieties of tomatoes bred in Siberia has been studied and summarized. As a result, the methods of genotyping tomato varieties and hybrids have been developed, where the main feature is the use of vertical electrophoresis in the 10% acrylamide gel. This method features high sensitivity and allows separating the fragments of different length into few nucleotide pairs. It is accessible for most laboratories, since, unlike capillary electrophoresis traditionally used for fragmentary analysis, it does not require expensive equipment and highly specialized professionals. As a result, the authors presented a comparative analysis of 33 varieties of tomatoes bred in Siberia on the basis of which individual profiles of each variety and each hybrid have been formed, and their genetic certification has been performed.
