SUPPLEMENTATION OF L-CARNITINE ON EXPANSION OF CUMULUS CELLS AND LEVEL OF METAFASE II OF GOAT OOCYTE

Abstract

The aim of the study was to analyze L-Carnitine supplementation on In Vitro Maturation (IVM) medium for cumulus expansion and transformation of this goat oocyte. The study material was immature oocytes with compact cumulus cells resulting from aspiration of follicles of 2-6 mm diameter and IVM in 5% CO2 incubators, 95% humidity, 38.5 ° C for 26 hours. IVM medium is TCM 199 + FBS 10% + 1 μg/mL Oestradiol + 0.002 AU / mL FSH + L-Carnitine in several levels. The research design used was complete randomized design. Each treatment had 10 replications of sampling. If there is any difference between treatments then proceed with Duncan test. Evaluation of oocytes cultured by medium of TCM 199 + FBS 10% + 1 μg/mL Oestradiol + 0.002 AU / mL FSH + L-Carnitine in several levels: 0, 0.2,0.4, 0.6.0.8, 0.10, 1.2 and 1.4 mg/mL, showed a perfectly expanding cumulus cell of 62.5%, 62.74%, 67.78%, 68.42%, 70.1% 4.76.6%, 91.3%, 78.51% and the oocytes reached Metaphase II: 59.1%, 61.67%, 58.2%, 60.78 %, 66.4%, 76.23%, 88.71%, 77.5%. There was a difference in percentage of perfect cumulus cell expansion and metaphase-II levels at different levels of L-Carnitine supplementation (P <0.05). From the results of this study that the difference of LCarnitine concentration in IVM medium resulted in the expansion of perfect cumulus cells and different levels of metaphase II. Highest percentage of perfectly expelled cumulus oocyte cells and metaphase II at concentrations of L-Carnitine supplementation 1.2 mg/mL.
