GREEN SYNTHESIS OF ZNO NANOPARTICLES FROM OCIMUM SANCTUM LEAF EXTRACT AND EVALUATION OF THEIR ANTIBIOFILM ACTIVITY

Abstract

Researches towards harmful effects of Nanoparticles synthesized from chemical methods have led inclination towards green synthesis. This study was designed considering above in mind. Zinc oxide nanoparticles (ZnONPs) were synthesized by using plant extract of Ocimum sanctum and analyzed their effectiveness in prevention of biofilm formation by a gram positive (S.aureus), a gram negative (P.aeruginosa) and a fungus (C.albicans). The results were deduced in reference to positive and negative controls and also a biofilm forming strain of E.coli. Their effectiveness was observed between range of 200 μg/mL - 600 μg/mL and their minimum effective dosage could be determined by analyzing the MIC point of ZnONPs against the target organisms. The quantitative assay was done using tube method, quantification done by measuring absorbance on UV-Vis Double beam spectrophotometer in reference to positive and negative control. Characterization of ZnONP was done by Electron microscopy, XRD and UV spectra. The most effective antibiofilm effect was observed against P.aeruginosa (0.145±0.004) in acetone extract synthesized Nanoparticle and against fungus C.albicans (0.142±0.005) in nanoparticle synthesized by methanolic extract. Thus the study showed ZnONPs from Ocimum sanctum extract might play a significant role against prevention of biofilm formed by these microbes.
